| Description | Compared to other fluorescence-based (FLICA) analysis substrates or fluorescence inhibitors for Caspase, the substrate provided in the reagent detects Caspase-3/7 activity without inhibiting the apoptosis process in intact cells. The substrate consists of a fluorescent DNA dye coupled to the DEVD recognition sequence of Caspase-3/7. The substrate is initially non-fluorescent and enters the cytoplasm through the cell membrane. In apoptotic cells, Caspase-3/7 cleaves the substrate, releasing a high-affinity DNA dye that migrates to the cell nucleus, labels DNA, and emits bright green fluorescence. The substrate is dual-functional, capable of detecting Caspase-3/7 activity while also visualizing morphological changes in the nucleus during apoptosis. SuperView staining is compatible with formalin fixation and subsequent immunofluorescence experiments. |
