Description A dual staining kit combining Alcian Blue at pH 2.5 for acid mucopolysaccharide detection with the Periodic Acid-Schiff reaction for neutral mucopolysaccharide and glycogen visualization, enabling comprehensive carbohydrate histochemistry on tissue sections.
Intended Use Simultaneous visualization of acidic mucins (stained blue by Alcian Blue) and neutral mucins/glycogen (stained magenta by PAS reaction) in formalin-fixed paraffin-embedded tissue sections for pathological evaluation.
Principle / Technology Alcian Blue at pH 2.5 electrostatically binds to carboxylated and sulfated acid mucopolysaccharides; periodic acid oxidizes vicinal diols in neutral polysaccharides to aldehydes, which react with Schiff reagent to produce a magenta-colored complex.
Detection Method Bright-field light microscopy; acidic mucins appear blue, neutral mucins and glycogen appear magenta, mixed mucins show purple coloration
Sample Type Formalin-fixed paraffin-embedded tissue sections (4–5 μm thickness) on positively charged glass slides
