| Product Name | Alamar Blue Cell Viability Reagent (Resazurin Sodium Salt) |
| Catalog No. | CATR-HMM-0043 |
| Description | A non-toxic, cell-permeable redox indicator that uses the natural reducing power of living cells to convert the blue, non-fluorescent resazurin molecule to the pink, fluorescent resorufin. The fluorescence or color intensity is proportional to the number of metabolically active cells in culture. |
| Intended Use | Longitudinal monitoring of cell viability and proliferation without terminating the culture; suitable for continuous tracking of cell health in stem cell maintenance, 3D organoid culture, and extended drug treatment studies. |
| Principle / Technology | Resazurin (blue, weakly fluorescent) enters viable cells and is reduced by mitochondrial, cytoplasmic, and microsomal reductases (including NADH dehydrogenase and flavin reductases) to resorufin (pink, strongly fluorescent, Ex/Em: 560/590 nm). The degree of conversion is proportional to viable cell number. |
| Detection Method | Fluorescence measurement (Ex 530–570 nm, Em 580–620 nm) or colorimetric absorbance at 570 nm with reference at 600 nm using a microplate reader |
| Sample Type | Mammalian cells, bacterial and fungal cultures, 3D spheroid and organoid models, primary cells, stem cells; suitable for extended time-course experiments |
| Performance Range / Specifications | Linear detection: 100–50,000 cells per well; fluorescence development over 1–6 hours; signal stable for extended readings over days with reagent replacement |
| Sensitivity / LOD | Detection of approximately 50–100 viable cells per well in 96-well microplate format using fluorescence mode |
| Specificity | Resazurin reduction occurs in metabolically active cells of diverse origins; non-fluorescent background in cell-free medium is minimal; viable and non-viable cells are clearly discriminated by the degree of conversion |
| Reaction Conditions / Protocol | Add Alamar Blue reagent at 10% v/v directly to culture medium; incubate at 37°C with 5% CO2 for 1–4 hours; measure fluorescence (Ex/Em: 560/590 nm) or absorbance (570 nm); optionally continue culture after replacing reagent-containing medium with fresh medium |
| Components / Formulation | Sterile-filtered Alamar Blue reagent (resazurin sodium salt in phosphate-buffered saline, pH 7.4), supplied as 20× or 100× concentrate depending on formulation |
| Storage Conditions | 2–8°C protected from light; stable for 12 months; avoid repeated warming and cooling; do not freeze |
| Shelf Life | 12 months from date of manufacture |
| Package Specifications | 25 mL, 100 mL, 500 mL (supplied as 10× or ready-to-use formulation) |
| Product Form | Sterile ready-to-use or concentrated aqueous solution |
| Quality Control | Each lot tested for fluorescence generation kinetics with reference cell lines; sterility confirmed per USP standards; reduction efficiency monitored; endotoxin tested (<0.1 EU/mL) |
| Key Features | Non-toxic reagent permits repeated measurements on the same cell culture; cell-permeable for unbiased assessment; minimal sample manipulation; compatible with both fluorescence and absorbance plate readers; widely validated across thousands of publications |
