Description A non-toxic, cell-permeable redox indicator that uses the natural reducing power of living cells to convert the blue, non-fluorescent resazurin molecule to the pink, fluorescent resorufin. The fluorescence or color intensity is proportional to the number of metabolically active cells in culture.
Intended Use Longitudinal monitoring of cell viability and proliferation without terminating the culture; suitable for continuous tracking of cell health in stem cell maintenance, 3D organoid culture, and extended drug treatment studies.
Principle / Technology Resazurin (blue, weakly fluorescent) enters viable cells and is reduced by mitochondrial, cytoplasmic, and microsomal reductases (including NADH dehydrogenase and flavin reductases) to resorufin (pink, strongly fluorescent, Ex/Em: 560/590 nm). The degree of conversion is proportional to viable cell number.
Detection Method Fluorescence measurement (Ex 530–570 nm, Em 580–620 nm) or colorimetric absorbance at 570 nm with reference at 600 nm using a microplate reader

Reaction Conditions / Protocol Add Alamar Blue reagent at 10% v/v directly to culture medium; incubate at 37°C with 5% CO2 for 1–4 hours; measure fluorescence (Ex/Em: 560/590 nm) or absorbance (570 nm); optionally continue culture after replacing reagent-containing medium with fresh medium
