Description A concentrated protein loading buffer for SDS-PAGE containing SDS, glycerol, bromophenol blue tracking dye, and Tris-HCl buffer. The buffer denatures proteins and imparts negative charge for electrophoretic separation.
Intended Use Denaturation and preparation of protein samples for loading onto SDS-PAGE gels; ensures uniform migration and visualization of protein bands during electrophoresis.
Principle / Technology SDS binds to proteins in a constant mass ratio (~1.4 g SDS/g protein), imparting a uniform negative charge density that overwhelms intrinsic protein charge, enabling electrophoretic migration proportional to molecular weight.
Detection Method Electrophoretic separation on SDS-PAGE gel; visual tracking of migration front by bromophenol blue dye
