| Product Name | Urine Protein Content Assay Kit (Ponceau S, Colorimetric) |
| Catalog No. | REKI-0016 |
| Description | A complete colorimetric workflow for measuring protein in urine, an indicator whose persistent elevation accompanies kidney injury and disease progression. Ponceau S binds urinary proteins to form an insoluble dye-protein complex that is pelleted by centrifugation, dissolved in alkaline buffer and read at 560 nm against a BSA standard curve. A sulfosalicylic acid turbidity pre-screen guides sample dilution before quantification, and a microplate reader can replace the cuvette spectrophotometer. Each 100-test pack supplies sulfosalicylic acid solution, 10x Ponceau S reagent, alkaline buffer, BSA standard and standard diluent. |
| Intended Use | Quantification of total protein in urine for nephropathy research and kidney injury studies |
| Principle / Technology | Ponceau S forms an insoluble complex with urinary protein; the dissolved pellet is measured at 560 nm, absorbance being proportional to protein concentration |
| Sample Type | Fresh urine specimens |
| Specificity | 100-test kit; five components (sulfosalicylic acid solution 100 mL, 10x Ponceau S reagent 10 mL, alkaline buffer 200 mL, BSA standard 20 mg, standard diluent 1.5 mL); working range 0-1600 ug/mL standards |
| Reaction Conditions / Protocol | Pre-screen with sulfosalicylic acid, dilute per the reference table, precipitate with 1x Ponceau S working solution, dissolve the pellet in alkaline buffer and read at 560 nm |
| Storage Conditions | Room temperature, protected from light |
| Package Specifications | 100 tests |
| Key Features | Integrated semi-quantitative pre-screen; spectrophotometer or microplate reader compatible |
| Readouts / Downstream Assays | Absorbance at 560 nm (540-595 nm acceptable) |
For research use only, not for clinical use.
