Product Name TUNEL Apoptosis Detection Kit (Fluor 640)
Catalog No. RESK-0171
Description When cells undergo apoptosis, endogenous DNA endonucleases are activated and cleave genomic DNA between nucleosomes; DNA extracted from apoptotic cells shows a characteristic 180-200 bp DNA ladder upon electrophoresis. The TUNEL Apoptosis Detection Kit (Fluor 640) is used to detect nuclear DNA fragmentation during late-stage apoptosis in tissue and cultured cells. Its principle is that, under the action of Terminal Deoxynucleotidyl Transferase (TdT), Fluor 640-12-dUTP is incorporated at the 3'-hydroxyl (3'-OH) termini exposed at genomic DNA breaks, allowing detection by fluorescence microscopy or flow cytometry. Fluor 640 is a red fluorescent dye with very high stability and strong brightness.
Intended Use Detection of nuclear DNA fragmentation in late-stage apoptosis of tissue and cultured cells.
Principle / Technology TUNEL (TdT-mediated dUTP nick end labeling). Terminal Deoxynucleotidyl Transferase (TdT) catalyzes the incorporation of Fluor 640-12-dUTP at the 3'-hydroxyl (3'-OH) termini exposed at genomic DNA breaks generated during apoptosis.
Detection Method Fluorescence microscopy or flow cytometry (red fluorescence).
Sample Type Frozen or paraffin-embedded tissue sections; cultured adherent or suspension cells.
Performance Range / Specifications Broad applicability: suitable for detecting apoptosis in frozen or paraffin-embedded tissue sections and in cultured adherent or suspension cells. Fluor 640 is a red fluorescent dye with very high stability and strong brightness.
Reaction Conditions / Protocol When combining TUNEL staining with immunofluorescence, perform the TUNEL reaction first and then the immunofluorescence staining, because immunofluorescence procedures may affect the TdT enzyme.
Immediate detection after staining is not mandatory; samples can be kept if stored protected from light.
Components / Formulation Fluor 640-12-dUTP Labeling Mix
Terminal Deoxynucleotidyl Transferase (TdT)
Storage Conditions Store the kit at -20 °C. The Fluor 640-12-dUTP Labeling Mix should be stored at -20 °C protected from light.
Shelf Life 1 year
Package Specifications 20 T
50 T
100 T
Product Form Kit
Shipping Conditions Shipped on wet ice.
Expiration Date / Stability Stable for 1 year when stored as recommended.
Regulatory / Compliance For research use only.
Compatibility Can be combined with immunofluorescence staining (perform TUNEL first, then immunofluorescence).
Application Notes / Precautions Diffuse DNA staining with poor nuclear localization or apparent cytoplasmic signal indicates non-specific staining, which may result from excessive staining time or overly high reagent concentration; reduce the staining solution concentration or shorten the staining time.
Store stained samples protected from light if immediate detection is not possible.

For research use only, not for clinical use.
