| Product Name | Hoechst 33342/PI Double Stain Kit |
| Catalog No. | RESK-0197 |
| Description | The Hoechst 33342/PI Double Stain Kit uses dual staining with Hoechst 33342 and propidium iodide (PI) to distinguish apoptotic cells from necrotic cells. Hoechst 33342 is a nuclear fluorescent dye that penetrates the cell membrane and emits blue fluorescence upon intercalating into double-stranded DNA. It enters normal cells only to a limited extent, producing weak blue staining. Because the membrane permeability of apoptotic cells is increased, substantially more Hoechst 33342 enters apoptotic cells than normal cells, resulting in stronger fluorescence. In addition, chromatin condensation during apoptosis allows the dye to bind DNA more efficiently and in a more concentrated manner, and the impaired p-glycoprotein pump function of the apoptotic cell membrane fails to effectively export Hoechst 33342, further increasing its intracellular accumulation. Together these features make the fluorescence of stained apoptotic cells markedly stronger than that of normal cells. PI cannot penetrate normal or apoptotic cells with intact cell membranes, so live cells are refractory to PI staining. Necrotic cells, however, lose membrane integrity at an early stage and can be stained by PI. After dual staining with these two dyes, detection by flow cytometry or fluorescence microscopy gives the following patterns: Normal cells: weak red fluorescence + weak blue fluorescence Apoptotic cells: weak red fluorescence + strong blue fluorescence Necrotic cells: strong red fluorescence + weak blue fluorescence |
| Intended Use | Differentiation of normal, apoptotic, and necrotic cells by dual nuclear staining with Hoechst 33342 and propidium iodide (PI), detected by flow cytometry or fluorescence microscopy. |
| Principle / Technology | Hoechst 33342 crosses the cell membrane and intercalates into double-stranded DNA to emit blue fluorescence; apoptotic cells show enhanced blue fluorescence owing to increased membrane permeability, chromatin condensation, and impaired p-glycoprotein-mediated dye efflux. PI is membrane-impermeant and only stains necrotic cells that have lost membrane integrity, emitting red fluorescence. |
| Detection Method | Flow cytometry or fluorescence microscopy. |
| Sample Type | Cultured cells; each sample can contain 10^5-10^6 cells. |
| Performance Range / Specifications | The kit is sufficient for 100 samples, with up to 10^5-10^6 cells per sample. Staining concentrations: Hoechst 33342 at 1 mg/mL; PI at 200 μg/mL. |
| Specificity | Hoechst 33342 stains the DNA of both normal and apoptotic cells, with markedly stronger fluorescence in apoptotic cells; PI selectively stains necrotic cells that have lost membrane integrity and is excluded by live and apoptotic cells. |
| Reaction Conditions / Protocol | Staining is fast and convenient and can be performed by a one-step or two-step method. For flow cytometric detection, no dilution or other preparation steps are needed and no additional solutions are required. Staining with Hoechst 33342 at 37 °C is recommended; staining at 4 °C may result in incomplete nuclear staining. |
| Components / Formulation | Hoechst 33342 staining solution (1 mg/mL) and propidium iodide (PI) staining solution (200 μg/mL). |
| Storage Conditions | Store at 4 °C for up to 12 months, or at -20 °C for up to 24 months. Aliquoting is recommended to avoid repeated freeze-thaw cycles. |
| Shelf Life | 12 months at 4 °C; 24 months at -20 °C. |
| Package Specifications | 1 Kit (100 tests) |
| Product Form | Liquid staining solutions. |
| Concentration | Hoechst 33342: 1 mg/mL; PI: 200 μg/mL. |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Stable for 12 months at 4 °C or 24 months at -20 °C; aliquot to avoid repeated freeze-thaw cycles. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Compatible with flow cytometry and fluorescence microscopy. |
| Application Notes / Precautions | Result interpretation after dual staining: Normal cells: weak red fluorescence + weak blue fluorescence Apoptotic cells: weak red fluorescence + strong blue fluorescence Necrotic cells: strong red fluorescence + weak blue fluorescence Staining with Hoechst 33342 at 37 °C is recommended, because staining at 4 °C may lead to incomplete nuclear staining. |
