Product Introduction Succinate dehydrogenase (SDH), or Complex II of the mitochondrial electron transport chain (ETC), is a unique enzyme that participates in both the tricarboxylic acid (TCA) cycle and oxidative phosphorylation. It catalyzes the oxidation of succinate to fumarate, transferring electrons to ubiquinone. Its activity is a direct indicator of mitochondrial integrity and function. This kit measures SDH activity by tracking the reduction of a water-soluble electron acceptor, which is coupled to a colorimetric probe, providing a sensitive readout suitable for tissue homogenates and isolated mitochondria.
Product Application • Assessment of mitochondrial function and bioenergetic capacity in cells and tissues.
• Research on mitochondrial disorders, neurodegenerative diseases, and ischemia-reperfusion injury.
• Toxicology studies evaluating drug or chemical-induced mitochondrial dysfunction.
• Investigation of the role of SDH mutations (e.g., in paragangliomas) or its role in metabolic signaling.
Product Advantages • Mitochondrial-Specific: Assay conditions are optimized to reflect the activity of the intact mitochondrial complex, minimizing non-specific signals.
• No Interference from NADH: The assay is independent of NADH/NAD+ redox status, allowing clear isolation of Complex II activity.
• Sensitive Detection: Can detect activity changes in small sample amounts, such as biopsy specimens or primary cell cultures.
• Functional Insight: Provides a direct measure of a key enzymatic step linking metabolism to energy production.
