Description 0.1 M sodium citrate buffer solution, pH 3.0, prepared from trisodium citrate dihydrate and citric acid. The citrate buffer system (pKa₁ 3.13, pKa₂ 4.76, pKa₃ 6.40) provides effective buffering across three pH ranges. At pH 3.0, this buffer is near the first pKa, ideal for acid elution of antibodies in affinity chromatography, antigen retrieval for immunohistochemistry at low pH, and pepsin digestion protocols.
Intended Use Low-pH antibody elution from Protein A/G affinity columns; antigen retrieval for IHC (low-pH citrate method); pepsin and acid protease digestion; RNA hydrolysis; zymography renaturation buffer component; heavy metal chelation in biochemical assays.
Principle / Technology Citric acid/sodium citrate buffer system; pKa₁ = 3.13 provides buffering at pH 2.2-3.8; citrate is also a mild chelating agent for divalent cations (Ca²⁺, Mg²⁺); pH 3.0 is below the isoelectric point of most antibodies causing elution from Protein A/G
Detection Method Ready-to-use at 0.1 M; further dilute as needed; for antibody elution: apply to Protein A/G column, incubate 2-5 min, collect fractions into 1/10 volume neutralization buffer (1 M Tris, pH 8.5)
