Product Name RIPA Lysis Buffer, 1x, Ready-to-Use
Catalog No. ATR-B-YJL-0178
Description A complete radioimmunoprecipitation assay lysis buffer containing ionic and non-ionic detergents with protease and phosphatase inhibitors for efficient extraction of total cellular proteins.
Intended Use Extraction of total protein from cultured cells and tissue samples for Western blotting, immunoprecipitation, and protein quantification assays.
Principle / Technology Combination of sodium deoxycholate (ionic), NP-40 (non-ionic), and SDS (denaturing) detergents disrupts cellular membranes and solubilizes cytoplasmic, membrane, and nuclear proteins.
Detection Method BCA or Bradford protein quantification, Western blot analysis of specific markers (cytoplasmic GAPDH, membrane EGFR, nuclear histone H3)
Sample Type Mammalian cultured cells, fresh and frozen tissue biopsies
Performance Range / Specifications Protein yield 2–5 mg/mL from confluent 10 cm dish of HeLa cells
Sensitivity / LOD N/A
Specificity Extracts cytoplasmic, membrane, nuclear, and cytoskeletal protein fractions
Reaction Conditions / Protocol Add cold RIPA buffer to washed cell monolayer or homogenized tissue; incubate on ice 15–30 minutes with intermittent vortexing; centrifuge at 14,000 × g for 15 minutes at 4°C; collect supernatant.
Components / Formulation Tris-HCl pH 7.4 50 mM, NaCl 150 mM, NP-40 1%, sodium deoxycholate 0.5%, SDS 0.1%, EDTA 1 mM, protease inhibitor cocktail, phosphatase inhibitor cocktail
Storage Conditions 2–8°C, protect from light
Shelf Life 12 months; use within 3 months after adding inhibitors
Package Specifications 100 mL, 500 mL
Product Form Liquid, ready to use (inhibitors pre-added)
Quality Control pH verification, lysis efficiency comparison with reference HeLa lysate, protease and phosphatase inhibitor activity verified
Key Features Complete formulation with inhibitors included; extracts total cellular protein; validated for IP and Western blot; compatible with protein assays
