| Product Name | Red Blood Cell Lysis Buffer |
| Catalog No. | RESK-0513 |
| Description | This Red Blood Cell Lysis Buffer provides a simple and easy-to-use method for removing red blood cells from fresh blood of humans, mice, rats, and other mammals. The optimized formulation fully lyses and removes red blood cells without damaging leukocytes, and typically recovers more than 80% of the total leukocytes in the blood sample while fully preserving leukocyte biological activity. The main component of this product is ammonium chloride. It is supplied as a ready-to-use solution that rapidly and efficiently lyses red blood cells and enriches leukocytes. The recovered leukocytes show good biological activity and can be used directly in downstream experiments such as flow cytometry, nucleic acid or protein extraction, and PCR. |
| Intended Use | For lysis of red blood cells and enrichment/isolation of leukocytes from fresh mammalian whole blood (human, mouse, rat, and other mammals). The recovered leukocytes are suitable for flow cytometry, nucleic acid or protein extraction, PCR, and other downstream experiments. |
| Principle / Technology | Ammonium chloride-based selective lysis of red blood cells while leaving leukocytes intact. |
| Sample Type | Fresh mammalian whole blood (human, mouse, rat, and other mammals). |
| Performance Range / Specifications | Typically recovers more than 80% of total leukocytes from the blood sample while fully preserving leukocyte biological activity. |
| Reaction Conditions / Protocol | 1. Equilibrate the refrigerated Red Blood Cell Lysis Buffer to room temperature before use. 2. Add 3 volumes of lysis buffer to the blood sample (e.g., add 900 uL of lysis buffer to 300 uL of blood). Mix by inversion and let stand at room temperature for 5 min, inverting to mix several times during the incubation. 3. Centrifuge at 10,000 rpm for 1 min (if the maximum speed of the centrifuge does not allow this, centrifuge at 3,000 rpm for 5 min). Aspirate and discard the supernatant. 4. Wash once with an appropriate buffer (e.g., PBS, pH 7.2-7.4). 5. After centrifugation, retain the leukocyte pellet for subsequent procedures. |
| Components / Formulation | Ammonium chloride-based, ready-to-use solution. Sterile and formaldehyde-free. |
| Storage Conditions | Store at 2-8 °C. Short-term storage at room temperature is also possible. |
| Shelf Life | 1 year when stored at 2-8 °C; 3 months when stored at room temperature. |
| Package Specifications | 100 mL / 250 mL / 500 mL |
| Product Form | Ready-to-use sterile liquid solution |
| Quality Control | Sterile. |
| Regulatory / Compliance | For research use only. |
| Compatibility | The recovered leukocytes can be used directly in flow cytometry, nucleic acid or protein extraction, PCR, and other downstream experiments. |
| Recommended Buffer System | Wash the cells once with PBS (pH 7.2-7.4) after lysis. |
| Application Notes / Precautions | Equilibrate the lysis buffer to room temperature before use, as the hemolytic efficiency of the lysis buffer is affected by temperature. This buffer is sterile and contains no formaldehyde. Leukocytes obtained after lysis are suitable for downstream nucleic acid extraction, PCR, and other experiments. No test data are available on whether the use of this reagent in flow cytometry affects the detection of circulating tumor cells (CTCs) in blood. |
For research use only, not for clinical use.
