| Reaction Conditions / Protocol | 1. Before use, allow the reagents to equilibrate to room temperature. 2. Before opening the cytokine vial cap, gently tap the vial or centrifuge it (10,000-12,000 rpm for 30 s or 3,000-3,500 rpm for 5 min) so that any small amount of lyophilized powder adhering to the vial wall or the inside of the cap settles to the bottom. 3. Disinfect the vial cap with a 75% alcohol cotton ball, then open the cap slowly and carefully to avoid powder loss caused by airflow disturbance. 4. Before reconstitution, refer to the "Reconstitution" section of the corresponding cytokine product manual to confirm the appropriate reconstitution solution and diluent. 5. According to the cytokine manual, add an appropriate volume of cytokine reconstitution solution (the reconstitution solution in this kit is Water For Injection for cell culture), and dissolve thoroughly by gentle pipetting or low-speed shaking to prepare a high-concentration stock solution; a concentration of 0.1-1.0 mg/mL is recommended. Note: Do not use a vortex mixer or other instruments for rapid shaking, to avoid bubble formation. 6. The reconstituted cytokine can be stored at 2-8 °C for up to 1 week. For long-term storage, dilute it to an appropriate concentration with the cytokine diluent (the diluent in this kit is PBS-BSA solution), mix gently by pipetting, aliquot according to single-use amounts, and freeze at -25 to -15 °C or -85 to -75 °C; avoid repeated freeze-thaw cycles. Note: A final concentration of no less than 10 μg/mL is recommended. 7. If a further reduction of cytokine concentration is required before adding the cytokine to the culture medium, perform serial dilution with the cytokine diluent (the diluent in this kit is PBS-BSA solution). Direct dilution of more than 500-fold is not recommended, in order to reduce pipetting error and ensure homogeneous mixing. Notes: 1. Low-concentration working solutions should be prepared fresh before use. If temporary storage is necessary, keep at 2-8 °C for no more than 24 hours (cytokine activity may decrease by 10-20% during storage). 2. Low-protein-binding centrifuge tubes and pipette tips are recommended to reduce protein loss caused by nonspecific adsorption. |
