| Product Name | Permeabilization Wash Buffer |
| Catalog No. | RESK-0514 |
| Description | Fixative and permeabilization reagents are used to treat the cell membrane before intracellular cytokine staining. The fixative stabilizes the cell membrane and maintains the binding of cell-surface antibodies to their antigens, while the permeabilization buffer creates small pores in the fluid, intact cell membrane to facilitate antibody entry into the cell. When intracellular antigens and DNA are detected by flow cytometry, the test cells are fixed and permeabilized, the corresponding antibodies or detection reagents are added, and after incubation for a defined period the samples can be analyzed on the instrument. |
| Intended Use | For permeabilization of cell membranes prior to intracellular cytokine staining and flow cytometric detection of intracellular antigens and DNA, in combination with a fixative. |
| Principle / Technology | Saponin-based, reversible permeabilization of the cell membrane, generating small pores that allow antibodies to enter the cell. |
| Detection Method | Flow cytometry |
| Sample Type | Test cells for intracellular antigen, intracellular cytokine, or DNA staining. |
| Reaction Conditions / Protocol | Application example (for reference only): 1. Harvest the test cells, centrifuge at 1,000 rpm for 10 min, and discard the supernatant. 2. Add 500 uL of pre-cooled (4 °C) fixative. 3. Incubate at room temperature for 10 min. 4. Centrifuge at 1,000 rpm for 10 min. 5. Discard the supernatant, wash once with PBS, and centrifuge at 1,000 rpm for 10 min. 6. Discard the supernatant and add 1.5 mL of permeabilization buffer. 7. Incubate at room temperature for 10-15 min. 8. Centrifuge at 1,000 rpm for 5 min. 9. Discard the supernatant and resuspend the cells in an appropriate amount of permeabilization buffer or flow cytometry staining buffer. 10. Add the detection antibody and stain for 15 min. 11. Wash once with PBS and centrifuge at 1,000 rpm for 5 min. 12. Discard the supernatant, resuspend the cells in 200 uL of PBS, and analyze by flow cytometry. |
| Components / Formulation | Saponin-based permeabilization buffer. |
| Storage Conditions | Store at 2-8 °C. Avoid direct exposure to strong light. Do not freeze. |
| Shelf Life | 1 year. |
| Package Specifications | 150 mL |
| Product Form | Liquid |
| Regulatory / Compliance | For research use only. |
| Compatibility | For immunofluorescence applications, this permeabilization buffer can be used to prepare the blocking solution (formulated with 1% BSA), and can also be used as the antibody dilution buffer and the wash buffer. |
| Recommended Buffer System | PBS for washing; permeabilization buffer or flow cytometry staining buffer for cell resuspension. |
| Application Notes / Precautions | Saponin is a relatively ideal cell permeabilization agent, but it cannot permeabilize the nuclear membrane. For flow cytometric analysis of nuclear factors, Triton X-100 and Tween-20 should be used if nuclear membrane permeabilization is required. The recommended staining sequence is fixation first, then permeabilization, then staining; surface and intracellular targets need to be stained together after permeabilization. Permeabilization with this buffer is reversible. |
For research use only, not for clinical use.
