| Description | The One-Step Mycoplasma Detection Kit is a rapid diagnostic product developed using isothermal amplification technology to detect mycoplasma contamination in cell culture media. The principle behind the test is that if a cell culture is contaminated with mycoplasma, the conserved sequences of mycoplasma DNA are amplified rapidly and in large quantities, causing the reaction solution to change from blue-violet to sky blue. The results are visually discernible and do not require electrophoresis. This kit can detect a wide range of mycoplasmas, including the eight species commonly found in cell culture, and its sensitivity and accuracy far exceed those of PCR methods. |

| Sample Preparation | Adherent cells: Directly aspirate the supernatant. It is recommended to collect samples at least 3 days after cell passaging or medium change, when confluence reaches approximately 90%. At this stage, the concentration of mycoplasma in the supernatant is higher, facilitating detection. Suspension cells: Centrifuge at 500 g for 5 minutes, then aspirate the supernatant. It is recommended to collect samples at least 3 days after cell passaging or medium change, as mycoplasma levels are higher at this time, facilitating detection. |
| Test Procedure | Remove Reagent from -20 °C, dissolve it, invert the tube to mix thoroughly, and centrifuge briefly to ensure all liquid settles at the bottom of the tube. Test Sample: Add 1 μL of the culture supernatant to the remaining reaction tubes; Negative Control: Do not add any sample to the first reaction tube; this serves as the negative control; Positive Control: Add 1 μL of the Positive Control to the last reaction tube; this serves as the positive control. After sample addition, allow the samples to stand at 30 °C for 5 minutes, then incubate at a constant temperature of 63 °C for 60 minutes in a water bath or PCR machine. |
| Results | Immediately after the 60-minute reaction at 63 °C, remove the reaction tubes and allow them to cool to room temperature. Observe the reaction results in a well-lit environment (a white background is recommended). If the reaction solution remains blue-violet, the result is negative; if the reaction solution is sky blue, the result is positive. |
| Storage and Transportation | Ship with ice packs. Store at -20 °C away from light. Shelf life: 18 months. If not using for an extended period, be sure to store away from light. |
