| Product Name | Mouse Tissue Direct PCR Kit (With Dye) |
| Catalog No. | RESK-0056 |
| Description | This kit performs rapid direct PCR amplification on mouse tissue samples (such as tail, ear, toe and muscle) with exceptional sample compatibility. A powerful lysis buffer quickly lyses samples and releases genomic DNA, and the lysate can be added directly into the PCR reaction without purification. Sample input requirements are minimal: 5 mg of mouse tissue or 1-5 mm of mouse tail is sufficient. The supplied 2× Mouse Direct PCR Mix is a 2×-concentrated hot-start reaction solution containing every PCR component except template and primers, greatly simplifying setup and reducing contamination risk. A tracking dye is included, so PCR products can be loaded directly onto gels. The kit is ideal for transgenic identification and mouse genotyping. |
| Intended Use | Extraction-free direct PCR from mouse tissues for transgenic identification and mouse genotyping; lysis can be performed in 96-well plates for high-throughput workflows. |
| Principle / Technology | A two-reagent lysis system (lysis buffer plus lysis-termination buffer) releases genomic DNA from mouse tissue at 95°C for 10-30 min; samples need no cutting or grinding. The hot-start 2× Mouse Direct PCR Mix, with a premixed tracking dye, amplifies the target directly from the crude lysate without DNA purification. |
| Sample Type | Mouse tail, mouse ear, mouse toe, muscle; 5 mg of mouse tissue or 1-5 mm of mouse tail is sufficient. |
| Performance Range / Specifications | Template preparation completed in 10 min; PCR reaction completed in as little as 50 min Extension: 30 sec/kb; amplicon length up to 1 kb GC range: 30-65% No sample cutting or grinding required for lysis Low material consumption; lysis can be performed in 96-well plates for high throughput |
| Specificity | More complete gene release than conventional alkaline lysis, verified by direct amplification of the SOX21 gene from mouse tail lysates. |
| Reaction Conditions / Protocol | Add lysis Component A and lyse the tissue at 95°C for 10-30 min; then add stop Component B separately and mix to terminate lysis. Centrifuge and use the supernatant (optional); no purification required. Recommended annealing temperature: Tm - (2-5)°C; extension 30 sec/kb; 35 cycles typical. Hot-start formulation. PCR products can be loaded directly onto gels thanks to the premixed tracking dye. |
| Components / Formulation | Component A: Buffer ML (lysis buffer; store at 2-8°C) Component B: Buffer MT (lysis-termination buffer; store at -25 to -15°C, avoid repeated freeze-thaw) Component C: 2× Mouse Direct PCR Mix, hot-start, with tracking dye (store at -25 to -15°C, avoid repeated freeze-thaw) |
| Storage Conditions | Component A (Buffer ML): store at 2-8°C; aliquot and freeze for long-term storage to avoid cross-contamination. Component B (Buffer MT): store at -25 to -15°C; avoid repeated freeze-thaw cycles. Component C (2× Mouse Direct PCR Mix): store at -25 to -15°C; avoid repeated freeze-thaw cycles. |
| Shelf Life | 1 year when stored as directed. |
| Package Specifications | 50 T 200 T |
| Product Form | Kit |
| Concentration | 2× (PCR mix) |
| Expiration Date / Stability | 1 year kit validity when components are stored as directed. |
| Compatibility | Compatible with mouse tail, ear, toe and muscle tissues; tracking dye premixed for direct gel loading after PCR. |
| Application Notes / Precautions | Lysis is performed at 95°C for 10-30 min; lysis Component A and stop Component B are added separately, and mixing after Component B terminates lysis immediately. No purification is needed; centrifugation to collect the supernatant is optional. Mouse tail is the most commonly used sample. PCR products can be loaded directly onto gels without adding loading buffer. |
