Product Name Mouse Tissue Direct PCR Kit Pro
Catalog No. RESK-0054
Description This kit combines rapid tissue lysis reagents with a high-performance amplification mix, allowing mouse tissue samples (such as tail, ear, toe, liver and kidney) to be lysed directly for fast release of genomic DNA. It is also compatible with whole blood and cell suspension samples. The lysate supernatant can be added straight into the PCR reaction without any purification, making the workflow exceptionally convenient. Sample input requirements are minimal: 2-5 mm² of mouse ear or liver, 1-5 mm of mouse tail, or 1-2 toes are sufficient. The supplied 2× Tissue Direct PCR Mix (With Dye) is a 2×-concentrated reaction solution containing every PCR component except template and primers, greatly simplifying setup, reducing contamination risk, and reliably amplifying fragments up to 10 kb. The kit is ideal for transgenic identification and mouse genotyping.
Intended Use Extraction-free direct PCR from mouse tissues for transgenic identification, mouse genotyping, and related research applications; also supports direct amplification from whole blood and cell suspensions.
Principle / Technology A rapid lysis reagent releases genomic DNA directly from mouse tissue (95°C for 3 min or 60°C for 15 min). The inhibitor-tolerant 2× Tissue Direct PCR Mix (with premixed blue tracking dye) accepts lysate at up to 20% of the reaction volume and amplifies targets up to 10 kb at 5-10 sec/kb extension, with no DNA purification required.
Sample Type Mouse tail, mouse ear, mouse toe, liver, kidney; also compatible with whole blood and cell suspensions. Minimal input: 2-5 mm² mouse ear or liver, 1-5 mm mouse tail, or 1-2 toes.
Performance Range / Specifications Direct amplification of fragments up to 10 kb
Fast extension: 5-10 sec/kb
High inhibitor tolerance: lysate input up to 20% of the PCR reaction volume
Flexible lysis: 95°C for 3 min or 60°C for 15 min
Multiplex amplification: up to 5-7 targets (500 bp-5 kb in 7-plex)
GC coverage: 30-70%
Lysate stability: amplification maintained after 4 weeks at 25°C or 4°C
Kit stability: amplification maintained after 4 weeks at 25°C/4°C or 2 weeks at 37°C
Specificity Reliable amplification of targets from 1 to 10 kb across 30-70% GC content directly from mouse tissue lysates.
Reaction Conditions / Protocol Lyse tissue at 95°C for 3 min (or 60°C for 15 min); add lysate supernatant directly to the PCR reaction (up to 20% of reaction volume; 1 μL lysate per 20-25 μL reaction is typical).
Extension: 5-10 sec/kb. Hot-start formulation.
For whole blood or cell samples, lysis can be omitted; extend the pre-denaturation step to 3 min.
Blue loading dye is premixed, so PCR products can be loaded directly onto gels.
Components / Formulation Component A: tissue lysis reagent (store at 4°C after opening)
Component B: tissue lysis reagent (store at 4°C after opening)
Component C: 2× Tissue Direct PCR Mix (With Dye), containing hot-start polymerase, dNTPs, Mg2+, buffer and blue tracking dye (store at -25 to -15°C after opening)
Storage Conditions Unopened kit: store at -25 to -15°C.
After opening: Components A and B at 4°C; aliquot if used repeatedly over long periods to avoid cross-contamination.
Component C at -25 to -15°C; avoid repeated freeze-thaw cycles.
Shelf Life 1 year (unopened and after opening, when stored as directed).

Product Form Kit
Concentration 2× (PCR mix)
Expiration Date / Stability 1 year validity; lysates remain amplifiable after 4 weeks at 25°C or 4°C, and the full kit remains functional after 4 weeks at 25°C/4°C or 2 weeks at 37°C.
Compatibility Compatible with mouse tail, ear, toe, liver and kidney tissues, as well as whole blood and cell suspensions; blue loading dye premixed for direct gel loading.
Application Notes / Precautions Whole blood or cell samples can be amplified directly without lysis; extend pre-denaturation to 3 min.
No additional loading buffer is required; the blue loading dye is already premixed into the PCR mix.
Aliquot components if used repeatedly over extended periods to avoid cross-contamination, and avoid repeated freeze-thaw of Component C.

For research use only, not for clinical use.
