| Assay Principle | Leucine arylamidase (LAP) is an enzyme that specifically hydrolyzes peptides containing N-terminal leucine residues. It is abundantly expressed in various tissues—particularly the liver, as well as the kidney and pancreas. The assay principle involves the enzymatic cleavage of the substrate L-Leucine-p-nitroanilide, releasing p-nitroaniline, which exhibits strong absorbance at 405 nm. The activity of LAP is determined by monitoring the rate of increase in absorbance at this wavelength, providing a direct and quantitative measure of enzymatic activity. |
