| Product Introduction | Alpha-ketoglutarate (α-KG) is a central metabolite in the tricarboxylic acid (TCA) cycle, a key nitrogen carrier in amino acid metabolism, and an essential cofactor for α-KG-dependent dioxygenases, including epigenetic regulators like TET enzymes and histone demethylases. This kit measures α-KG concentration through a coupled enzymatic reaction where α-KG is converted to L-glutamate by glutamate dehydrogenase (GLDH) with the concomitant oxidation of NADH. The decrease in NADH absorbance at 340 nm is directly proportional to the α-KG content. |
| Product Application | • Research on mitochondrial function, TCA cycle flux, and cellular bioenergetics. • Studies in cancer metabolism, immunometabolism, and aging. • Investigation of amino acid metabolism (transamination) and nitrogen balance. • Epigenetics research focused on α-KG-dependent enzyme activity (e.g., in stem cell biology). |
| Product Advantages | • High Specificity via GLDH: Utilizes the specificity of glutamate dehydrogenase for α-KG, ensuring minimal cross-reactivity with other keto acids like pyruvate or oxaloacetate. • Sensitive Kinetic Assay: The kinetic measurement of NADH oxidation provides high sensitivity and a wide linear range, suitable for low-concentration samples like cell culture media or tissue extracts. • Direct & Simple Protocol: Requires only sample deproteinization followed by the enzymatic reaction, offering a straightforward path to accurate results without complex derivatization. • Microplate-Optimized: Configured for 96-well plates, enabling high-throughput analysis of many samples, which is ideal for screening experiments or large-scale metabolic phenotyping. |
For research use only, not for clinical use.
