Description Transfusion reactions caused by irregular antibodies are getting more and more attention, and the problem of the production of homologous antibodies caused by incompatible transfusion is especially prominent in patients with thalassemia, sickle cell anemia and other long-term transfusion.PCR genotyping technology is simple and rapid, and is a better complementary method to the current serological detection technology. In addition, in fetal hemolytic disease of the newborn (HDNF) transfusion therapy, 70-80% of the homologous antibodies in the recipient's serum are related to RhCcEe antibodies. The risk of HDNF is predicted by testing the parental RhCcEe gene and predicting the fetal RhCcEe genotype. If necessary, amniotic fluid can be tested (or a noninvasive fetal free RhCcEe gene test kit can be used) for final confirmation of fetal blood group.
Applications This kit can qualitatively detect the intron 2 insertion 109bp, c.307T>C, c.308C>T, c.676C>G, c.733C>G, RHCE*Exon8 of the RHCE gene in DNA samples to predict the RhCE blood group, and provide a reference for the identification of the RhCE blood group.
Sample Type Whole blood ≥1 mL (sodium citrate or EDTA anticoagulation); DNA ≥10 μL (concentration 20-40 ng/μL, A260/A280 value 1.6-2.0); Amniotic fluid: ≥4 mL (more than 16 weeks), the above three kinds of specimens were stored at 2-8°C for 2 weeks, and ≤-20°C for 1 year. Oral mucosa swabs (2, respectively, with brushing force to collect oral mucosa cells from both cheeks, stored at room temperature for 1 week).

Estimated Measurement Time 1 hour and 41 minutes (one-step (10min) operation, amplification products do not need to be transferred for spiking, avoiding laboratory and aerosol contamination).
