Product Name Exosome Lysis Buffer
Catalog No. RESK-0616
Description This lysis buffer is a lysis reagent developed specifically for exosome samples. It efficiently lyses exosomes to release exosomal proteins, and it is pre-supplemented with protease inhibitors to effectively inhibit protein degradation. For use, simply mix with the exosome sample at a 1:1 ratio; the lysed protein sample can be used for BCA protein quantification, Western blot, and other experiments. This reagent contains a relatively high concentration of detergents and is not suitable for protein concentration determination by the Bradford method. Main components: Tris (pH 7.4), NaCl, Triton X-100, sodium deoxycholate, SDS, EDTA, PMSF, and a protease inhibitor cocktail (AEBSF, aprotinin, bestatin, E-64, leupeptin, pepstatin A).
Intended Use Efficient lysis of exosome samples for extraction of exosomal proteins for downstream BCA protein quantification and Western blot analysis. For research use only.
Principle / Technology Detergent-based lysis: a combination of ionic and non-ionic detergents (Triton X-100, sodium deoxycholate, SDS) disrupts the exosome lipid bilayer to release exosomal proteins, while PMSF and a protease inhibitor cocktail (AEBSF, aprotinin, bestatin, E-64, leupeptin, pepstatin A) inhibit protein degradation during lysis.
Detection Method Protein concentration determination by BCA assay and downstream Western blot analysis.
Sample Type Exosome samples.
Performance Range / Specifications Recommended mixing ratio: exosome sample to lysis buffer at 1:1 (v/v); lyse on ice for 10 min.
The protein concentration of the lysed sample should be less than 2 μg/uL to ensure complete lysis.
Demonstrated Western blot performance: the same exosome sample lysed with different reagents was quantified and analyzed by Western blot (10% SDS-PAGE separating gel, 10 μg loading, CD63 primary antibody 1:2000 incubated overnight at 4 °C, mouse IgG secondary antibody 1:10000 incubated 50 min at room temperature, 5 s exposure).
Reaction Conditions / Protocol 1. Before the experiment, thaw the lysis buffer at 2-8 °C, mix well, and place on ice.
2. Mix the exosome sample with the lysis buffer at a 1:1 volume ratio and lyse on ice for 10 min.
3. Centrifuge at 11,200 x g (approximately 12,000 rpm on a microcentrifuge with an effective radius of about 7 cm, for centrifuge tubes up to 2 mL) for 5 min at 4 °C and collect the supernatant.
4. Determine the protein concentration by BCA assay and proceed to Western blot or other downstream experiments.

Compatibility Lysed protein samples are suitable for BCA protein quantification and Western blot. Due to the relatively high detergent concentration, this buffer is not suitable for protein concentration determination by the Bradford method.
Application Notes / Precautions This product contains protease inhibitors; avoid repeated freeze-thaw cycles, and aliquoting for frozen storage is recommended.
All lysis steps must be performed on ice or at 4 °C.
The protein concentration of the lysed sample should be less than 2 μg/uL to ensure complete lysis.
