Description Alta DiagnoTech alpha-amylase reagent is a two-reagent EPS substrate system for quantitative in vitro measurement of alpha-amylase activity in human serum or plasma.
Intended Use In vitro quantitative determination of alpha-amylase activity in human serum or plasma; intended to support auxiliary assessment and treatment monitoring of acute pancreatitis.
Principle / Technology Alpha-amylase cleaves ethylidene-p-nitrophenyl-alpha-D-maltoheptaoside. Alpha-glucosidase then hydrolyzes the degradation products to p-nitrophenol, whose color intensity at 405 nm is proportional to alpha-amylase activity.
Detection Method Kinetic p-nitrophenol colorimetric assay at 405 nm
Sample Type Human serum or plasma
Components / Formulation Reagent 1: HEPES 50 mmol/L, sodium chloride 70 mmol/L, magnesium chloride 10 mmol/L and alpha-glucosidase >=4 kU/L. Reagent 2: EPS 3.5 mmol/L.
Storage Conditions Store unopened or opened reagent protected from light at 2-8 degrees C; do not freeze.
Shelf Life 12 months unopened when sealed and protected from light at 2-8 degrees C
Application Notes / Precautions Do not mix reagent lots; recalibrate when changing lots.
