| Description | A dye-binding kit for total protein quantification built on the Bradford reaction. When the Coomassie brilliant blue staining reagent binds protein under acidic conditions the absorbance maximum shifts immediately from 465 nm to 595 nm and the colour turns from brown to blue, and the protein concentration is calculated by comparing sample absorbance with a standard protein curve. A practical advantage of the method is that salts, solvents, metal chelators and buffers present in the sample have little influence on the measurement, so crude extracts and column fractions can be read with minimal preparation. |
