| Product Name | Annexin V-Fluor 647/7-AAD Apoptosis Detection Kit |
| Catalog No. | RESK-0173 |
| Description | The Annexin V-Fluor 647/7-AAD Apoptosis Detection Kit uses Annexin V labeled with the fluorescent dye Fluor 647 as a probe to detect the occurrence of early apoptosis in cells. Detection can be performed by fluorescence microscopy, flow cytometry, or other fluorescence detection equipment. The kit also provides 7-AAD, a nucleic acid dye that distinguishes live and early apoptotic cells from necrotic or late apoptotic cells, enabling clear discrimination of live cells (Annexin V-/7-AAD-), early apoptotic cells (Annexin V+/7-AAD-), and late apoptotic or necrotic cells (Annexin V+/7-AAD+). |
| Intended Use | Detection of early-stage apoptosis in cells by fluorescence-based analysis. |
| Principle / Technology | In normal viable cells, phosphatidylserine (PS) is located on the inner leaflet of the plasma membrane, but in early apoptotic cells PS translocates from the inner leaflet to the cell surface and becomes exposed to the extracellular environment. Annexin V is a Ca2+-dependent phospholipid-binding protein with a molecular weight of 35-36 kDa that binds PS with high affinity, allowing Fluor 647-labeled Annexin V to bind to the membrane of early apoptotic cells via the exposed phosphatidylserine. The 7-AAD provided in this kit distinguishes live and early apoptotic cells from necrotic or late apoptotic cells: 7-AAD is a nucleic acid dye with fluorescence properties similar to PI but a narrower emission spectrum, causing less interference with other detection channels and making it an ideal substitute for PI in multicolor fluorescence analysis. 7-AAD cannot penetrate the intact cell membranes of live or early apoptotic cells, but it can enter late apoptotic or necrotic cells and bind to their DNA. Therefore, when Fluor 647-labeled Annexin V is used together with 7-AAD, live cells are Annexin V-/7-AAD-, early apoptotic cells are Annexin V+/7-AAD-, and late apoptotic or necrotic cells are double-positive (Annexin V+/7-AAD+). |
