Product Name All-in-One RT-gDNA Digestion SuperMix for qPCR (with Red Tracking Dye)
Catalog No. RESK-0001
Description This upgraded one-tube, traceable reverse transcription reagent is designed specifically for qPCR applications. All components required for genomic DNA removal and reverse transcription are integrated into a single tube — simply add RNA template and RNase-free water to start the reaction. The formulation combines excellent tolerance to sample inhibitors, highly efficient genomic DNA elimination, and high-yield reverse transcription performance. A red tracking dye is built into the system, enabling visual pipetting control and reducing the risk of missed samples without affecting reverse transcription, genomic DNA digestion, or downstream qPCR. The resulting cDNA product is red; when added to a blue-dye qPCR master mix, the solution turns purple, providing an intuitive color change that confirms template addition. It can also be paired with other qPCR reagents, such as Alta DiagnoTech's instrument-universal qPCR master mixes, to achieve pipetting visualization, although the color transition may vary slightly between different qPCR reagents.
Intended Use One-tube genomic DNA removal and reverse transcription of total RNA to generate cDNA for real-time quantitative PCR (RT-qPCR) analysis.
Principle / Technology Single-tube supermix integrating a genomic DNA digestion step with reverse transcriptase-mediated cDNA synthesis; a built-in red tracking dye provides visual pipetting control without interfering with enzymatic performance or downstream qPCR detection.
Detection Method Downstream real-time quantitative PCR (qPCR) analysis of the synthesized cDNA
Sample Type Total RNA (validated on human, mouse, and Arabidopsis RNA samples)
Performance Range / Specifications Genomic DNA removal and reverse transcription are completed in a single 15-min program. In parallel comparisons with mainstream comparable products using 100 ng of human, Arabidopsis, and mouse RNA as templates, this reagent delivered lower Ct values, indicating higher reverse transcription efficiency, with a particularly marked advantage on mouse RNA templates.
Sensitivity / LOD High reverse transcription efficiency and strong tolerance to sample inhibitors, yielding lower Ct values than comparable products in side-by-side testing.
Reaction Conditions / Protocol Recommended single program: 37°C for 10 min (genomic DNA removal) followed by 60°C for 5 min (reverse transcription); total reaction time 15 min. If needed, an alternative condition of 50°C for 5-15 min may be tested to confirm that reverse transcription efficiency meets experimental requirements.
Components / Formulation All-in-one RT-gDNA digestion supermix (single tube) containing all components required for genomic DNA removal and reverse transcription, including a red tracking dye.
Only RNA template and RNase-free water need to be added by the user.
Storage Conditions Store at -25°C to -15°C
Shelf Life 1 year
Package Specifications 100 reactions (100 T)
Product Form All-in-one premix (supermix)
Expiration Date / Stability Stable for 1 year when stored at -25°C to -15°C.
Compatibility The red cDNA product can be combined with Alta DiagnoTech's blue-dye qPCR SYBR Green master mixes for full RT-qPCR pipetting tracking (red + blue turns purple), and is also compatible with other qPCR reagents, including instrument-universal qPCR mixes; color transitions may differ slightly between reagents. In principle it can be used with qPCR mixes from other suppliers, but performance should be verified before use.
Application Notes / Precautions This is a qPCR-dedicated reverse transcription reagent; it is not recommended for conventional PCR amplification. For PCR applications, use Alta DiagnoTech's first-strand cDNA synthesis kits.
The recommended program (37°C, 10 min; 60°C, 5 min) provides optimal gDNA removal and reverse transcription efficiency; 50°C for 5-15 min may be evaluated if required.
Pairing with a blue-dye qPCR master mix produces a red-to-purple color change for visual confirmation of template addition.
Compatibility with qPCR mixes from other suppliers has not been validated; test performance before use.
