Product Name Advanced One-Step RT-qPCR SYBR Green Kit
Catalog No. RESK-0066
Description The Advanced One-Step RT-qPCR SYBR Green Kit performs fluorescence-based quantification using SYBR Green I dye. With gene-specific primers, reverse transcription and qPCR are completed in a single tube, eliminating repeated cap-opening and pipetting steps, greatly improving detection efficiency and reducing contamination risk. For RNA samples, a thermostable reverse transcriptase efficiently synthesizes cDNA, while an antibody-based hot-start Taq DNA polymerase drives quantitative amplification. Within its optimized buffer system, the kit achieves detection sensitivity down to 0.1 pg for highly expressed targets and 1 pg for moderately expressed targets. The kit is equally suitable for amplification and quantification of DNA samples, enabling highly sensitive detection and quantification of nucleic acids from a wide range of plant and animal samples, cells, and microorganisms.
Intended Use Highly sensitive one-step RT-qPCR detection and quantification of RNA (and direct qPCR of DNA) from plant and animal samples, cells, and microorganisms.
Principle / Technology One-tube reverse transcription and qPCR with gene-specific primers; thermostable reverse transcriptase for efficient cDNA synthesis combined with an antibody-based hot-start Taq DNA polymerase in an optimized buffer system.
Detection Method SYBR Green I dye-based fluorescence detection.
Sample Type RNA samples from plants, animals, cells, and microorganisms; also suitable for direct amplification and quantification of DNA samples.
Performance Range / Specifications Universal instrument compatibility: suitable for common real-time PCR instruments of all ROX types (No ROX, Low ROX, High ROX); targets of varying GC content (30%-70%) are efficiently amplified with single, sharp melt peaks.
High sensitivity: down to 0.1 pg for highly expressed RNA targets and 1 pg for moderately expressed targets; viral RNA detectable to the E2 level; DNA templates detectable down to single-copy level.
Excellent repeatability: minimal variation across 90 replicate wells at high, medium, and low template inputs.
High precision: accurately discriminates 2-fold differences in RNA or DNA template concentration.
Excellent stability: assembled reaction mixes can stand at 4°C for 24 h without performance change; reagents show no significant difference after 20 h of ambient light exposure.
Annealing temperature 60°C; amplicon length 80-300 bp.
Sensitivity / LOD RNA targets: 0.1 pg (high expression), 1 pg (moderate expression); viral RNA to the E2 level. DNA targets: single-copy detection with amplification signal.
Specificity Single melt peaks across targets of varying GC content (30%-70%); hot-start Taq polymerase suppresses nonspecific amplification.
Reaction Conditions / Protocol Reverse transcription and qPCR are performed sequentially in the same tube; recommended annealing temperature 60°C; recommended amplicon length 80-300 bp. For DNA samples, the reverse transcription step can be skipped and qPCR performed directly.

Product Form Mix
Quality Control Sensitivity, repeatability, precision, and stability verified across multiple instruments and template types; 2-fold concentration discrimination confirmed by 2-fold serial dilution of 293T cell RNA.
Expiration Date / Stability 1 year at -25°C to -15°C protected from light; assembled reaction mixes stable at 4°C for 24 h.
Compatibility Compatible with the vast majority of real-time PCR instruments, including No ROX and Low/High ROX platforms; universal ROX formulation. GC range 30%-70%.
Recommended Buffer System 2x optimized SYBR Green buffer with dNTPs (included).
Application Notes / Precautions Suitable for RNA or DNA detection from plant and animal samples, cells, and microorganisms; for RNA samples the kit efficiently synthesizes cDNA and performs quantitative amplification, while DNA samples can be amplified directly without a reverse transcription step.
Store at -25°C to -15°C protected from light; valid for 1 year.
Supports No ROX and Low/High ROX instruments; refer to the manual for specific models.
Batch-to-Batch Consistency Minimal variation across 90 replicate wells at high, medium, and low template inputs demonstrates strong reproducibility.
