Product Introduction ADP-glucose pyrophosphorylase (AGPase) is the rate-limiting enzyme in starch biosynthesis in plants and glycogen synthesis in bacteria. It catalyzes the reaction: ATP + α-Glucose-1-P ↔ ADP-Glucose + PPi. This enzyme is a major regulatory point, controlling carbon flux into storage polymers, and is a target for crop yield improvement. This kit measures AGPase activity in the reverse (pyrophosphorolysis) direction by coupling the production of ATP to a highly sensitive bioluminescent detection system, offering superior sensitivity over traditional methods.
Product Application • Fundamental research on starch metabolism in leaves, seeds, tubers, and other storage organs.
• Genetic engineering and screening of plant varieties for altered starch content or quality.
• Studies of bacterial glycogen metabolism and its role in persistence and virulence.
• Biochemical characterization of AGPase enzyme kinetics and allosteric regulation.
Product Advantages • High-Sensitivity Luminescence: The luciferase-based detection provides exceptional sensitivity, ideal for low-activity samples or small tissue amounts.
• Specific Reverse Reaction: Measuring pyrophosphorolysis minimizes interference from other ATP-consuming or producing reactions in crude extracts.
• Optimized for Plant Tissues: Includes recommendations for preparing active enzyme extracts from challenging plant materials.
• High-Throughput Screening: The microplate format is perfectly suited for mutant screening or inhibitor studies.
