| Product Name | 2× Ultra-Rapid Hot-Start PCR Master Mix (With Dye) |
| Catalog No. | RESK-0047 |
| Description | The 2× Ultra-Rapid Hot-Start PCR Master Mix (With Dye) is built on an engineered Taq DNA polymerase, fortified with a potent elongation factor, an amplification enhancer, and an optimized buffer system for exceptionally high amplification efficiency. Complex templates such as genomic DNA up to 3 kb can be extended at 1-3 sec/kb, and simple templates such as plasmids up to 5 kb at 1 sec/kb, substantially reducing total PCR run time. The ready-to-use 2× format contains dNTPs and Mg2+, so only primers and template need to be added, greatly simplifying the workflow. An electrophoresis tracking dye is premixed into the master mix, allowing PCR products to be loaded directly onto gels. Built-in protectants keep the enzyme fully active after repeated freeze-thaw cycles, and the resulting PCR products carry 3'-dA overhangs for effortless cloning into T vectors. |
| Intended Use | Ultra-rapid PCR and gene identification across a broad range of species. |
| Principle / Technology | Hot-start amplification based on an engineered Taq DNA polymerase, combined with a potent elongation factor, an amplification enhancer, and an optimized buffer system. |
| Sample Type | Genomic DNA (including human genomic DNA), plasmid DNA, and other templates with 30-70% GC content. |
| Performance Range / Specifications | Extension speed: 1-3 sec/kb for complex templates (genomic DNA) up to 3 kb; 1 sec/kb for simple templates (plasmids) up to 5 kb. Amplicon length: up to 15 kb overall; up to 8 kb for genomic DNA. GC compatibility: 30-70% GC content. Annealing temperature: 60°C or 1-2°C below the primer Tm. |
| Sensitivity / LOD | High amplification sensitivity, demonstrated by sensitivity testing on an 8 kb human genomic DNA target. |
| Specificity | Strong amplification specificity with high detection rates; validated on human genomic DNA targets of 1, 2, 3, 4, and 8 kb. |
| Reaction Conditions / Protocol | Annealing temperature: 60°C or 1-2°C below the primer Tm. Extension time: 1-10 sec/kb depending on template type and amplicon length. Add only primers and template to the 2× mix. |
| Components / Formulation | Engineered hot-start Taq DNA polymerase, dNTPs, Mg2+, elongation factor, amplification enhancer, optimized buffer system, electrophoresis tracking dye, and stabilizing protectants. |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | 2 years when stored at -25°C to -15°C. |
| Package Specifications | 1 mL; 5× 1 mL; 50× 1 mL; 100× 1 mL |
| Product Form | Premixed master mix (Mix) |
| Quality Control | Performance verified by amplification of human genomic DNA targets from 1 to 8 kb; stability confirmed after storage at 37°C for up to 14 days across templates with 30-70% GC content. |
| Concentration | 2× |
| Source / Origin | Engineered Taq DNA polymerase. |
| Expiration Date / Stability | Stable for 2 years at -25°C to -15°C. Performance remains stable after 14 days at 37°C, and the mix tolerates repeated freeze-thaw cycles. |
| Compatibility | Suitable for templates with 30-70% GC content; PCR products carry 3'-dA overhangs compatible with T-vector (TA) cloning; contains a tracking dye for direct gel loading. |
| Application Notes / Precautions | Only primers and template need to be added; no additional dNTPs or Mg2+ required. The mix contains an electrophoresis tracking dye, allowing direct gel loading after PCR. PCR products carry 3'-dA overhangs and can be cloned directly into T vectors. The formulation tolerates repeated freeze-thaw cycles without loss of activity. |
| Batch-to-Batch Consistency | Stable amplification performance maintained after 14 days at 37°C and across templates with 30-70% GC content. |
