| Product Name | 2× Robust PCR Master Mix (With Dye) |
| Catalog No. | RESK-0051 |
| Description | This 2× Robust PCR Master Mix (With Dye) is built on an engineered Taq DNA polymerase combined with a powerful processivity-enhancing factor, an amplification-boosting factor, and a fully optimized buffer system, delivering a step-change in both speed and yield over conventional PCR mixes. Extension rates of 15 sec/kb are routinely achieved for fast PCR, and amplicons within 1 kb can be extended at an extreme rate of 5 sec/kb, dramatically shortening run times. The premix already contains dNTPs and Mg2+, so only primers and template need to be added, greatly simplifying setup. An integrated electrophoresis tracking dye allows the reaction to be loaded directly onto a gel after PCR. Protective agents in the formulation keep the master mix active through repeated freeze-thaw cycles. PCR products carry a 3'-dA overhang and can be cloned directly into T vectors. |
| Intended Use | Routine and fast PCR amplification and gene identification/genotyping from purified DNA templates, including animal, plant and human genomic DNA, bacterial colonies, and plasmid DNA. |
| Principle / Technology | An engineered Taq DNA polymerase is combined with a potent extension-enhancing factor, an amplification-enhancing factor and an optimized buffer containing dNTPs and Mg2+. A gel-tracking dye is premixed into the 2× formulation, and stabilizing agents preserve enzyme activity after repeated freeze-thaw cycles. Amplification products carry 3'-dA overhangs compatible with TA cloning. |
| Sample Type | Human genomic DNA, Arabidopsis genomic DNA, rice genomic DNA, mouse genomic DNA, bacterial cultures (colony PCR), plasmid DNA. |
| Performance Range / Specifications | Extension rate: 15-30 sec/kb; extreme rate of 5 sec/kb for targets within 1 kb Amplicon length: 1-4 kb (up to 5 kb) Broad template compatibility (animal, plant and human gDNA, colony PCR, plasmid DNA) Target GC tolerance: 35-70% |
| Specificity | Recommended cycling conditions resolve the majority of targets; if non-specific amplification occurs, increasing the annealing temperature in 2°C steps is recommended. |
| Reaction Conditions / Protocol | Add primers and template directly to the 2× master mix. Recommended annealing temperature: 55°C (adjust within 1-2°C). Extension: 15-30 sec/kb depending on amplicon length; load the reaction directly onto an agarose gel after PCR. |

| Concentration | 2× |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Stable for 2 years at -25 to -15°C; the formulation remains active after repeated freeze-thaw cycles. |
| Compatibility | PCR products carry a 3'-dA overhang for direct TA cloning. Suitable for agarose gel electrophoresis; not suitable for PAGE. For crude templates (e.g., mouse tail or plant lysates), a hot-start PCR genotyping master mix is recommended instead. |
| Application Notes / Precautions | Shipped with ice packs; store at -20°C for a 2-year shelf life. PCR products from this mix are not suitable for PAGE analysis. For crude templates such as mouse tail or plant lysates, use a hot-start genotyping master mix. If non-specific bands appear, raise the annealing temperature in 2°C increments. |
For research use only, not for clinical use.
