Product Name 2× High-Fidelity PCR Master Mix
Catalog No. RESK-0036
Description This product is a ready-to-use 2× premixed solution containing a high-fidelity DNA polymerase, dNTPs, and an optimized buffer system. The included enzyme is a new-generation high-fidelity DNA polymerase with 5'→3' DNA polymerase activity and 3'→5' exonuclease (proofreading) activity, delivering a fidelity 52-fold higher than Taq DNA polymerase and 6-fold higher than conventional Pfu DNA polymerase, with extension rates of up to 15 sec/kb. It is suitable for the amplification of complex templates and produces blunt-ended products. The master mix is fast, convenient, highly sensitive, specific, and stable: only primers and template need to be added to set up the reaction, simplifying workflows, reducing pipetting errors, and increasing throughput and reproducibility. This product contains no loading dye, so loading buffer must be added to PCR products before electrophoresis. A specific protective agent keeps the premix active even after repeated freeze-thaw cycles.
Intended Use High-fidelity PCR amplification for gene cloning, site-directed mutagenesis, and accurate amplification of complex templates where low error rates are required.
Principle / Technology Based on a proofreading high-fidelity DNA polymerase with 3'→5' exonuclease activity that corrects misincorporated bases during 5'→3' DNA synthesis, achieving a fidelity 52× that of Taq DNA polymerase.
Sample Type DNA templates including complex templates; verified amplification up to 20 kb for simple templates and 10 kb for complex templates.
Performance Range / Specifications Fidelity: 52× Taq DNA polymerase and 6× conventional Pfu DNA polymerase; error rate approximately 0.00039% (Taq: ~0.0002, about one mutation per 5 kb)
Extension rate: up to 15 sec/kb
Amplification length: verified up to 20 kb for simple templates and 10 kb for complex templates
Product ends: blunt ends
Format: 2× ready-to-use premix; only primers and template required
Stability: retains stable activity after repeated freeze-thaw cycles
Specificity High amplification specificity from the proofreading high-fidelity polymerase in an optimized buffer system.
Reaction Conditions / Protocol 2× ready-to-use format: add only primers and template to the reaction. Extension rate up to 15 sec/kb. Amplification products are blunt-ended and require loading buffer before gel electrophoresis (no dye in the mix).
Components / Formulation 2× ready-to-use premixed solution containing high-fidelity DNA polymerase, dNTPs, and an optimized buffer system; contains a specific protective agent for freeze-thaw stability; contains no loading dye.

Expiration Date / Stability Contains a specific protective agent; the premix maintains stable activity after repeated freeze-thaw cycles. Valid for 2 years when stored at -20°C.
Compatibility Suitable for amplification of complex templates; blunt-ended products are compatible with downstream cloning workflows.
Recommended Buffer System Built-in optimized buffer system supplied at 2× concentration.
Application Notes / Precautions Fidelity is based on 3'→5' exonuclease proofreading activity; error rate is approximately 0.00039% (52× higher fidelity than Taq). Higher fidelity reduces but cannot completely eliminate mutations.
Products are blunt-ended. To add A-tails for TA cloning: purify the PCR product (≥20 ng/μL), combine 14.5 μL of product with 2 μL Taq buffer, 3 μL dNTPs, and 0.5 μL Taq polymerase, incubate at 72°C for 10–20 min, then purify.
Targets within the verified range (up to 20 kb for simple templates, 10 kb for complex templates) are theoretically amplifiable; actual results depend on template and primer quality.
If mutations appear in products, check whether the original template carries mutations or has degraded from repeated freeze-thaw cycles or prolonged UV exposure; use a higher-fidelity enzyme if PCR-introduced mutations are suspected.
Batch-to-Batch Consistency The premixed format reduces manual pipetting errors and improves experimental throughput and result reproducibility.
