| Product Name | 2× Antibody Hot-Start PCR Master Mix (With Dye) |
| Catalog No. | RESK-0073 |
| Description | 2× Antibody Hot-Start PCR Master Mix (With Dye) is a ready-to-use PCR premix containing antibody hot-start Taq DNA polymerase, dNTPs and an optimized buffer system; only primers and template need to be added, greatly simplifying reaction setup and improving throughput and reproducibility. The hot-start Taq polymerase is a mixture of a high-affinity anti-Taq antibody and Taq DNA polymerase: the antibody keeps polymerase activity blocked even after 30 minutes at 50°C, and a 30-second heating step at the pre-denaturation temperature fully inactivates the antibody to release polymerase activity, effectively suppressing non-specific amplification from primer mis-annealing. The premix already contains electrophoresis buffer and a tracking dye, so PCR products can be loaded directly onto a gel; the dye does not interfere with amplification efficiency. PCR products carry 3'-dA overhangs and can be cloned directly into T-vectors. |
| Intended Use | For low-copy-number gene amplification, genotyping and colony PCR. |
| Principle / Technology | Antibody-mediated hot start: a high-affinity anti-Taq antibody blocks Taq DNA polymerase activity during reaction setup (blocking maintained even at 50°C for 30 min); a 30 sec heating step at the pre-denaturation temperature inactivates the antibody and releases polymerase activity, preventing non-specific amplification. |
| Detection Method | PCR amplification; products contain dye and electrophoresis buffer for direct agarose gel electrophoresis. |
| Performance Range / Specifications | Suitable for low-copy gene amplification, genotyping and colony PCR. 2× ready-to-use premix: add only primers and template. Contains tracking dye and electrophoresis buffer for direct gel loading; the dye does not affect amplification efficiency. PCR products carry 3'-dA overhangs for direct T-vector cloning. |
| Specificity | Antibody hot-start mechanism effectively suppresses non-specific amplification caused by non-specific primer annealing. |
| Reaction Conditions / Protocol | Add primers and template to the 2× master mix; after a brief pre-denaturation step (30 sec antibody inactivation), run a standard PCR program. Products can be loaded directly onto agarose gels without adding loading buffer. |
| Components / Formulation | 2× ready-to-use master mix containing antibody hot-start Taq DNA polymerase, dNTPs, optimized buffer, electrophoresis buffer and tracking dye. |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | 2 years when stored at -25°C to -15°C. |
| Package Specifications | 100 μL; 1 mL; 5× 1 mL |
| Product Form | Liquid 2× premix (with dye). |
| Concentration | 2× |
| Application Notes / Precautions | Fidelity is comparable to conventional wild-type Taq DNA polymerase. The hot-start mechanism avoids non-specific amplification during reaction setup. No additional loading buffer is required before gel electrophoresis; the dye does not interfere with amplification efficiency. |
