Product Name 1st Strand cDNA Synthesis SuperMix II
Catalog No. RESK-0028
Description This ready-to-use premix contains a second-generation thermostable reverse transcriptase, RNase inhibitor, dNTPs, a Random Primers/Oligo dT primer mix, and an optimized buffer system; only template RNA and RNase-free H2O need to be added to run the reaction. The second-generation reverse transcriptase is an engineered enzyme derived from M-MLV (RNase H-) reverse transcriptase. Compared with M-MLV (RNase H-) reverse transcriptase, its thermostability is greatly improved, tolerating reaction temperatures up to 50°C, which makes it suitable for reverse transcribing RNA templates with complex secondary structures. The enzyme also shows enhanced template affinity, making it well suited for low-input templates and low-copy-number genes.
Intended Use First-strand cDNA synthesis from RNA templates; the reverse transcription product is intended primarily for downstream qPCR. For research use.
Principle / Technology The second-generation reverse transcriptase is a novel enzyme obtained by genetic engineering of M-MLV (RNase H-) reverse transcriptase. It offers greatly improved thermostability (tolerating reaction temperatures up to 50°C) for reverse transcription of RNA templates with complex secondary structures, together with enhanced template affinity for low-input templates and low-copy-number genes.
Detection Method Downstream qPCR of the synthesized cDNA (short-fragment PCR is also possible).
Sample Type Template RNA. The species origin of the RNA has little impact on reverse transcription; RNA quality is the key factor, and RNA from any species, including fungi, can be used.
Performance Range / Specifications Ready-to-use premix format; tolerates reverse transcription reaction temperatures up to 50°C.
Sensitivity / LOD Enhanced template affinity makes the premix suitable for low-input templates and low-copy-number genes.
Reaction Conditions / Protocol Add template RNA and RNase-free H2O to the ready-to-use premix to set up the reaction; reverse transcription can be performed at temperatures up to 50°C for RNA templates with complex secondary structures.

Second-generation reverse transcriptase
RNase inhibitor
dNTPs
Random Primers/Oligo dT primer mix
Optimized buffer system

Source / Origin The reverse transcriptase is an engineered enzyme derived from M-MLV (RNase H-) reverse transcriptase.
Shipping Conditions Shipped with ice packs.
Expiration Date / Stability Stable for 18 months when stored at -20°C.
Application Notes / Precautions Primer selection guide: Oligo (dT) is recommended for full-length first-strand cDNA synthesis from eukaryotic templates; Random Primers are recommended when the target region has complex secondary structure or the template is of prokaryotic origin; gene-specific primers (GSP) suit known target sequences; for downstream qPCR, Oligo (dT) and Random Primers can be used as a mixture.
Polyadenylated lncRNA can be reverse transcribed with premix- or kit-format products; circRNA, miRNA, and non-polyadenylated lncRNA require kit-format products, and miRNA requires special stem-loop primers and a dedicated reverse transcription kit.
Cloning-oriented and quantification-oriented reverse transcription differ in purpose and process. cDNA from cloning-oriented RT can be used for qPCR, but cDNA from quantification-oriented RT (with mixed Oligo (dT)/Random Primers) is relatively short and is not recommended for conventional PCR of long fragments; only short-fragment PCR is feasible.
The species origin of the RNA has little impact on reverse transcription; RNA quality is the key factor.

For research use only, not for clinical use.
