Product Name 1st Strand cDNA Synthesis Kit II (with gDNA Digester)
Catalog No. RESK-0027
Description This first-strand cDNA synthesis kit incorporates a genomic DNA removal step and is developed on our second-generation thermostable reverse transcriptase. The enzyme offers greatly improved thermostability, tolerating reaction temperatures up to 50°C, which makes it suitable for reverse transcribing RNA templates with complex secondary structures. Enhanced template affinity supports low-input templates and low-copy-number genes, and the enzyme's ability to synthesize full-length cDNA is also improved, enabling amplification of cDNA up to 10 kb. The included gDNA digester removes residual genomic DNA contamination from RNA templates for more reliable downstream results. Two cDNA synthesis primers, Random Primers N6 and Oligo (dT)18, are provided in separate tubes; users can choose Random Primers N6, Oligo (dT)18, or gene-specific primers as the reverse transcription primer. The synthesized single-stranded cDNA can be used directly in downstream PCR or qPCR.
Intended Use First-strand cDNA synthesis with a genomic DNA removal step; the resulting single-stranded cDNA is suitable for direct use in downstream PCR or qPCR. For research use.
Principle / Technology Based on a second-generation thermostable reverse transcriptase that tolerates reaction temperatures up to 50°C and shows enhanced affinity for RNA templates, supporting reverse transcription of templates with complex secondary structures and of low-input or low-copy-number targets. The enzyme also delivers improved full-length cDNA synthesis, amplifying cDNA up to 10 kb. A supplied gDNA digester removes residual genomic DNA contamination from RNA templates.
Detection Method Downstream PCR or qPCR analysis of the synthesized cDNA.
Sample Type Template RNA, including total RNA and mRNA; also applicable to miRNA (stem-loop method), circRNA, and lncRNA (with or without poly-A tails).
Performance Range / Specifications Tolerates reverse transcription reaction temperatures up to 50°C; amplifies cDNA up to 10 kb.
Sensitivity / LOD Enhanced template affinity makes the kit suitable for low-input templates and low-copy-number genes. For miRNA reverse transcription, an input of 0.5-1 ug miRNA is recommended.
Specificity Two primers (Random Primers N6 and Oligo (dT)18) are supplied in separate tubes, and Random Primers N6, Oligo (dT)18, or gene-specific primers can be selected as the reverse transcription primer according to experimental needs.
Reaction Conditions / Protocol Perform gDNA digestion with the supplied gDNA digester and buffer, then carry out reverse transcription with the enzyme mix, buffer, and the primer of choice; reaction temperatures up to 50°C can be applied for templates with complex secondary structures.

Source / Origin Developed on a second-generation thermostable reverse transcriptase.
Shipping Conditions Shipped on dry ice.
Expiration Date / Stability Stable for 18 months when stored at -20°C.
Compatibility The synthesized single-stranded cDNA can be used directly in downstream PCR or qPCR.
Application Notes / Precautions For qPCR of lncRNA, add both Oligo (dT) and Random Primers, since some lncRNAs carry poly-A tails and others do not.
The kit can be used for miRNA reverse transcription by the stem-loop method; an input of 0.5-1 ug miRNA is recommended, as expression abundance varies among miRNAs.
The kit is suitable for reverse transcription of circRNA and non-polyadenylated lncRNA.
Kit-format products generate longer cDNA than SuperMix formats and are recommended when longer reverse transcription products are needed, such as for downstream plasmid construction.

For research use only, not for clinical use.
