Product Introduction NADP-malate dehydrogenase (NADP-MDH) is a key light-regulated enzyme in chloroplasts. In C4 plants, it is central to the CO₂ concentrating mechanism, reducing oxaloacetate to malate in the mesophyll cells. In C3 plants and other organisms, it functions in the malate valve, exporting reducing equivalents (as malate) from chloroplasts to the cytosol, thereby regulating the NADPH/NADP⁺ ratio and protecting against photo-oxidative stress. This kit measures NADP-MDH activity in the forward direction by monitoring the oxidation of NADPH at 340 nm, providing a direct and sensitive kinetic assay.
Product Application • Research on C4 photosynthesis, photosynthetic efficiency, and plant productivity.
• Studies of chloroplast redox regulation, photoprotection, and responses to light stress.
• Investigation of metabolic interactions between organelles (malate shuttle).
• Biochemical analysis of transgenic plants with altered C4 pathway enzymes.
Product Advantages • Direct Kinetic Measurement: UV-based monitoring of NADPH oxidation provides real-time, high-quality kinetic data.
• Chloroplast Enzyme Focus: Optimized for measuring activity in chloroplast extracts or leaf crude extracts where this enzyme is localized.
• High Specificity: The assay direction and conditions favor NADP-MDH over the more abundant NAD-dependent MDH isozyme.
• Reliable Performance: Yields highly reproducible activity values, crucial for comparative physiology studies.
