| Product Introduction | Cytoplasmic isocitrate dehydrogenase (ICDHc) catalyzes the oxidative decarboxylation of isocitrate to α-ketoglutarate, generating NADPH in the cytosol. Unlike its mitochondrial counterpart, ICDHc is not part of the TCA cycle but is a major source of NADPH for reductive biosynthesis (e.g., fatty acids, cholesterol) and for maintaining the glutathione-based antioxidant system. Its activity is linked to cell proliferation, cancer metabolism, and oxidative stress responses. This kit specifically measures the NADP⁺-dependent activity in the cytoplasmic fraction by monitoring NADPH production at 340 nm. |
| Product Application | • Research on cancer cell metabolism and the role of NADPH in supporting rapid growth and antioxidant defense. • Studies of lipogenesis in liver, adipose tissue, and other anabolic contexts. • Investigation of cellular responses to oxidative stress and the regulation of redox homeostasis. • Differentiation of metabolic roles between cytoplasmic and mitochondrial ICDH isoforms. |
| Product Advantages | • Compartment-Specific: The protocol includes guidance for preparing cytoplasmic extracts to specifically target the ICDHc isoform. • Sensitive UV Kinetic Assay: Provides continuous, high-sensitivity measurement of NADPH formation. • Specific for NADP⁺: Uses NADP⁺ as the cofactor, ensuring measurement of the biosynthetic/redox-regulating isoform. • High-Throughput Compatible: The 96-well UV-transparent plate format allows for efficient kinetic reading of multiple samples. |
