| Product Name | Exosome Protein Assay Kit for Western Blot (CD63 & TSG101) |
| Catalog No. | RESK-0200 |
| Description | The Exosome Protein Assay Kit for Western Blot (CD63 & TSG101) is a convenient and efficient kit developed for the detection of the exosome marker proteins CD63 and TSG101. The dedicated exosome protein lysis buffer included in the kit efficiently lyses exosomes to release exosomal proteins. The antibodies are strictly screened and validated for the detection of exosomal CD63 and TSG101 proteins, and the kit is suitable for exosome protein detection in human, rat and mouse samples. |
| Intended Use | Qualitative Western blot detection of the exosome marker proteins CD63 and TSG101 in exosome samples. |
| Principle / Technology | Exosomes are lysed with the dedicated lysis buffer; exosomal proteins are separated by SDS-PAGE and detected by Western blot using mouse monoclonal primary antibodies against CD63 and TSG101 and an HRP-conjugated goat anti-mouse IgG secondary antibody. |
| Detection Method | Western blot (SDS-PAGE followed by chemiluminescent detection with HRP-conjugated secondary antibody). |
| Sample Type | Exosome samples from human, mouse and rat. |
| Specificity | Anti-CD63 and Anti-TSG101 mouse monoclonal antibodies; applicable species: human, mouse, rat. |
| Reaction Conditions / Protocol | SDS-PAGE: 10% separating gel; sample loading: 20 μg protein per lane. Primary antibodies: Anti-CD63 antibody, 1:1000 dilution; Anti-TSG101 antibody, 1:1000 dilution; incubate overnight at 4 °C. Secondary antibody: Goat anti-Mouse IgG (HRP), 1:10000 dilution; incubate at room temperature for 50 min; exposure time 5 s. Lysis: add RIPA reagent at a 1:1 ratio to the exosome sample and lyse on ice for 10-15 min; perform BCA protein quantification before Western blot. |
| Components / Formulation | Exosome protein dedicated lysis buffer: 20 mL CD63 & TSG101 positive control: 50 μL (2 μg/μL) x 2 vials (prepared from serum exosome lysate; used to monitor the assay system) Anti-CD63 antibody (mouse monoclonal): 10 μL Anti-TSG101 antibody (mouse monoclonal): 10 μL Goat anti-Mouse IgG antibody (HRP): 10 μL |

| Expiration Date / Stability | 1 year when stored at -25 to -15 °C. Avoid repeated freeze-thaw cycles; aliquot according to single-use amounts. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Suitable for Western blot analysis of exosomes from human, mouse and rat samples; CD63 and TSG101 are the preferred markers for exosome identification. |
| Recommended Buffer System | Dedicated exosome protein lysis buffer (included); RIPA reagent (1:1 with sample) for lysis before Western blot. |
| Application Notes / Precautions | This is a qualitative detection; no loading-control (reference) protein is available for exosome Western blot identification. Avoid repeated freeze-thaw cycles; aliquot reagents according to single-use amounts. BCA and other Western blot related reagents must be supplied by the user. Perform BCA protein quantification before Western blot. Wear a lab coat and disposable gloves during operation. |
For research use only, not for clinical use.
