| Product Name | Exosome CD81 Protein Assay Kit |
| Catalog No. | RESK-0160 |
| Description | The Exosome CD81 Protein Assay Kit is a convenient and efficient kit developed for the detection of the exosome marker protein CD81. The exosome-specific protein lysis buffer included in the kit efficiently lyses exosomes to release exosomal proteins, while the CD81 positive control together with the primary and secondary antibodies allows monitoring of the experimental system and accurate detection of CD81 protein in exosomes. |
| Intended Use | Detection of the exosome marker protein CD81 in exosome samples by Western blot or ELISA. |
| Principle / Technology | Exosomes are efficiently lysed with the exosome-specific protein lysis buffer to release exosomal proteins, which are quantified (e.g., by BCA assay) and analyzed by Western blot or ELISA. The CD81 nanobody is a recombinant alpaca VHH fused to the rabbit IgG Fc domain; it detects a stronger, specific signal in exosome lysates with no reactivity toward other proteins. The CD81 positive control and the primary/secondary antibodies allow monitoring of the entire experimental system. |
| Detection Method | Western blot (HRP-based detection) and ELISA. |
| Sample Type | Exosome samples and exosome lysates (e.g., 293T cell-derived exosomes). |
| Performance Range / Specifications | The CD81 nanobody is a recombinant alpaca VHH fused with a rabbit IgG Fc domain. It specifically detects a stronger signal in exosome lysates, shows no reactivity with other proteins, and is suitable for both Western blot and ELISA detection. |
| Specificity | The CD81 nanobody (recombinant alpaca VHH fused to rabbit IgG Fc) specifically recognizes CD81 in exosome lysates with no cross-reactivity toward other proteins. |
| Reaction Conditions / Protocol | SDS-PAGE: 10% separating gel. Sample loading: positive control (PC) 10 uL; 293T cell exosomes 10 ug. Primary antibody: CD81 antibody, 1:1000 dilution, incubate overnight at 4 °C. Secondary antibody: Goat anti-Rabbit IgG, 1:2000 dilution, incubate 50 min at room temperature. |
| Components / Formulation | Exosome-specific protein lysis buffer, CD81 positive control, CD81 primary antibody (recombinant alpaca VHH fused to rabbit IgG Fc) and Goat anti-Rabbit IgG (HRP) secondary antibody. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 1 year |
| Package Specifications | 5 T |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Valid for 1 year when stored at -25 to -15 °C. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Suitable for Western blot and ELISA detection of CD81 in exosome samples. |
| Application Notes / Precautions | Before the experiment, thaw the lysis buffer at 2-8 °C, mix well and place on ice. Mix the exosome sample with the lysis buffer at a 1:1 volume ratio and lyse on ice for 10 min. Centrifuge at 12,000 x g for 5 min at 4 °C and collect the supernatant; determine protein concentration by BCA assay before Western blot or other downstream experiments. For Western blot, load 5-20 uL of the CD81 positive control in parallel with the experimental samples; when using the CD81 positive control for the first time, determine the appropriate loading volume in a preliminary experiment according to your own conditions. The recommended dilution of the Goat anti-Rabbit IgG (HRP) secondary antibody is 1:2000-1:5000. The CD81 positive control must be heat-denatured at 70 °C for 5 min before loading; reagents for the BCA assay and Western blot are not included and must be provided by the user. |
For research use only, not for clinical use.
