| Detection Principle | CISH technology can be used to detect and visualize specific nucleic acid sequences in cell preparations. CISH probes denature together with their complementary target sequences in the preparation, and then anneal and bind during hybridization. Subsequently, non-specific and unbound probe fragments are removed through rigorous washing steps. The double-stranded formation of labeled probes can be visualized using primary (unlabeled) antibodies, which are subsequently detected by secondary enzyme-labeled antibodies. The enzymatic reaction combines with a chromogenic substrate to form a colored precipitate. After counterstaining the cell nuclei with a nuclear dye, the hybridized probe fragments can be observed under a light microscope. |
