Product Name Caspase-8 Colorimetric Assay Kit
Catalog No. RESK-0181
Description Caspases are a family of proteases involved in apoptosis, comprising more than ten members. Caspase-8, also known as FLICE, MACH or Mch5, normally exists as a zymogen and is activated upon apoptosis; it is regarded as an upstream caspase in apoptotic signal transduction. During Fas-receptor- and TNFR-1-mediated apoptosis, caspase-8 is activated to form a dimer, which in turn activates the downstream caspase-4, -6, -9 and -10.
Intended Use For the detection of Caspase-8 activity in mammalian tissues and cells; suitable for studying Caspase-8 activity in apoptotic signaling pathways.
Principle / Technology The assay is based on the specific hydrolysis of the peptide substrate Ile-Glu-Thr-Asp-p-nitroanilide (IETD-pNA) by Caspase-8. The released free p-nitroaniline (pNA) shows maximum absorbance at 405 nm, and Caspase-8 activity is determined by colorimetric measurement of pNA.
Detection Method Colorimetric assay (visible spectrophotometry); absorbance read at 405 nm
Sample Type Mammalian tissues and cells
Performance Range / Specifications Caspase-8 activity is determined by visible-light colorimetric measurement of pNA. Suitable for the detection of Caspase-8 activity in mammalian tissues and cells.
Specificity Based on the specific hydrolysis of the IETD-pNA substrate by Caspase-8.
