Product Name Caspase-1 Colorimetric Assay Kit
Catalog No. RESK-0175
Description Caspases are a family of proteases involved in apoptosis, comprising more than ten members. Caspase-1 is the only caspase that cleaves the precursors of IL-1beta and IL-18 to generate the active cytokines. Caspase-11 cleaves the 45 kD pro-caspase-1 to produce 20 kD and 10 kD fragments, which form a heterodimer and further assemble into an enzymatically active tetramer. Caspase-1 regulates apoptosis by cleaving Bcl-XL and modulates cytokine-mediated immune and inflammatory responses through cleavage of cytokine precursors.
Intended Use For the detection of Caspase-1 activity in mammalian tissues and cells; suitable for apoptosis research.
Principle / Technology The assay is based on the specific hydrolysis of the peptide substrate Tyr-Val-Ala-Asp-p-nitroanilide (YVAD-pNA) by Caspase-1. The released free p-nitroaniline (pNA) shows maximum absorbance at 405 nm, and Caspase-1 activity is determined by colorimetric measurement of pNA.
Detection Method Colorimetric assay (visible spectrophotometry); absorbance read at 405 nm
Sample Type Mammalian tissues and cells
Performance Range / Specifications Caspase-1 activity is determined by visible-light colorimetric measurement of pNA. Suitable for the detection of Caspase-1 activity in mammalian tissues and cells.
Specificity Based on the specific hydrolysis of the YVAD-pNA substrate by Caspase-1.
