| Description | Microdeletions at the azoospermia factor (AZF) loci on the Y chromosome are a major genetic cause of male primary infertility. The AZF gene family consists of three regions: AZFa, AZFb, and AZFc; a microdeletion in any of these regions can lead to impaired spermatogenesis. This product utilizes multiplex PCR technology to rapidly detect six sequence-tagged sites (STS) within the AZFa, AZFb, and AZFc regions of the Y-chromosomal AZF gene family. Each sample requires two multiplex PCR reactions: Group A includes the sY254, sY127, and sY86 sites, while Group B includes the sY134, sY84, sY255. Each group uses two pairs of primers designed to amplify the ZFY and SRY genes as internal controls. |

| Test Procedure | Depending on laboratory practice, various methods may be used to extract genomic DNA from peripheral blood. Measure the concentration using a spectrophotometer, and finally dilute the DNA to 20–100 ng/μL for PCR amplification. PCR amplification: 94°C for 5 minutes; {94°C for 30 seconds; 55°C for 30 seconds; 65°C for 4 minutes; 25 cycles}; 65°C for 5 minutes; 4°C, store. Agarose gel electrophoresis: Directly load 10 μL of the PCR product onto a 3% agarose gel and run for 30–40 minutes. |
| Results | All male samples should amplify an internal control band. If this band is absent in the test sample, the experiment is considered a failure; in this case, verify whether the DNA extraction was successful or whether the amplification procedure was performed correctly. If one or more STS bands are missing compared to the normal control, it can be concluded that the corresponding region is deleted. |
