Description A rapid isothermal colorimetric assay for routine detection of mycoplasma contamination in cell-culture supernatants. The closed-tube workflow does not require PCR instrumentation or post-amplification electrophoresis.
Intended Use Routine screening of adherent and suspension cell cultures for mycoplasma contamination.
Principle / Technology Isothermal amplification generates a visual color change that indicates the presence of mycoplasma nucleic acid.
Detection Method Visual colorimetric endpoint after constant-temperature incubation.
Sample Type Cell-culture supernatant; serum can also be tested directly.
Performance Range / Specifications Uses 1 µL sample; detects up to 28 mycoplasma species, including eight commonly associated with cell-culture contamination.
Sensitivity / LOD At least 500 CFU in 1 µL culture supernatant, equivalent to approximately 5 × 10^5 CFU/mL.
Specificity Broad detection of common cell-culture mycoplasma with tolerance to culture-supernatant inhibitors.
Reaction Conditions / Protocol Add 1 µL supernatant to the reaction system, incubate at 60°C for 1 hour and interpret the color at the specified endpoint.
