| Description | The target antigen is coated onto a microplate to create a solid-phase carrier. Standards or samples are added to the wells, and the target binds to the antibody attached to the solid-phase carrier. Horseradish peroxidase-labeled antibody is then added. Unbound antibody is removed by washing, followed by thorough rinsing, and finally, TMB substrate is added to produce a color reaction. Under the catalysis of horseradish peroxidase, TMB is converted to blue, which is then converted to yellow under the action of acid. The intensity of the color is directly proportional to the concentration of the target in the sample. The absorbance (OD value) is measured at a wavelength of 450 nm using a microplate reader, and the sample concentration is calculated. |
