| Test Procedure | Combine a known volume (100 μL) of fluorescent beads with an equal volume of the test sample. Treat the sample with a lysis reagent to lyse and permeabilize cells, eliminating red blood cells and preparing the cells. Add a staining reagent containing propidium iodide and RNase. A flow cytometer measures the fluorescence emitted by each stained cell as it passes through the laser beam (since mature platelets and red blood cells lack DNA, the stained cells represent the white blood cell fraction). |
