| Product Name | Anti-DYKDDDDK Affinity Resin for Secretory Proteins (Clone 1A3, Mouse IgG2b) |
| Catalog No. | IVDRM-0287 |
| Description | Secreted proteins typically arrive in conditioned medium at modest concentration alongside a great deal of unrelated material, so capture specificity decides how clean the final preparation is. Clone 1A3 was selected for exactly this situation: immobilised on four percent highly cross-linked agarose, it pulls down Flag-tagged secreted targets with little non-specific binding. Capacity is at least 1.1 mg of protein per millilitre of settled resin. Binding accommodates the met-modified N-terminal arrangement as well as standard N-terminal and C-terminal Flag fusions, so construct design stays flexible, and release with competing Flag peptide keeps the recovered protein in native condition for downstream assays. |
| Intended Use | Purification and immunoprecipitation of Flag-tagged proteins secreted into culture medium. |
| Principle / Technology | Immobilised monoclonal antibody captures the DYKDDDDK epitope and the bound protein is displaced afterwards by soluble Flag peptide competing for the same site. |
| Sample Type | Conditioned culture medium and other fractions containing secreted Flag-tagged protein. |
| Performance Range / Specifications | Capacity at least 1.1 mg of protein per millilitre of resin; low non-specific binding; the monoclonal antibody is coupled at high density to cross-linked agarose; peptide-based elution preserves function. |
| Specificity | Mouse IgG2b clone 1A3 on four percent highly cross-linked agarose; capacity at least 1.1 mg per millilitre; supplied at 1 mL, 5 mL, 25 mL and 100 mL of settled resin; resin-to-protective solution ratio of one to one. |
| Reaction Conditions / Protocol | Equilibrate the resin, load conditioned medium, wash with Tris-buffered saline, elute with Flag peptide and desalt if the peptide interferes with downstream steps. |
| Components / Formulation | Settled resin suspended in protective solution. |
| Storage Conditions | Store at -25 to -15 °C; freezing outside the glycerol-containing solution is prohibited. |
| Shelf Life | 1 year from receipt when stored as recommended. |
| Package Specifications | Supplied as 1 mL, 5 mL, 25 mL or 100 mL of resin. |
| Product Form | Agarose resin suspension in protective solution. |
| Quality Control | Each lot is verified for clone identity and for capacity per millilitre of resin. |
| Key Features | - Clone 1A3 chosen for secreted protein capture - Capacity at least 1.1 mg of protein per millilitre of resin - Low non-specific binding even with abundant contaminating medium proteins - Accepts N-terminal, C-terminal and met-modified Flag constructs - Four resin volumes from 1 mL to 100 mL |
| Purity | Murine monoclonal antibody of the IgG2b subclass. |
| Concentration | At least 1.1 mg of protein bound per millilitre of resin. |
| Activity / Unit Definition | Measured as target captured per millilitre rather than as enzyme units. |
| Source / Origin | Murine hybridoma antibody immobilised on agarose. |
| pH Range / Optimal pH | Protective solution of Tris-buffered saline with fifty percent glycerol at pH 7.4. |
| Shipping Conditions | Shipped cold. |
| Expiration Date / Stability | Remains usable for the full shelf life provided the temperature recommendation is followed. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Suited to native elution workflows where the target must remain biologically active. |
| Recommended Buffer System | Tris-buffered saline with fifty percent glycerol at pH 7.4 and 0.02 percent mass per volume sodium azide. |
| Application Notes / Precautions | Keep the resin in its glycerol solution at all times, remove azide before work with living material, and count volume as settled resin rather than total suspension. |
| Batch-to-Batch Consistency | Lot-to-lot consistency is maintained through defined manufacturing specifications; every lot ships with its release record. |
| Model Type | Immunoaffinity capture resin |
| Starting Material / Tissue Site | Murine IgG2b clone 1A3 antibody immobilised on cross-linked agarose |
| Delivery Standard | Bottle of resin suspension in protective solution |
| Turnaround Time | dispatch is arranged at order confirmation. |
| Readouts / Downstream Assays | Recovery of the secreted target and its purity after elution as judged by SDS-PAGE. |
For in vitro diagnostic use.
