| Product Name | Anti-DYKDDDDK Affinity Resin for Secretory and Membrane Proteins (Clone 3F5, Mouse IgG2b) |
| Catalog No. | IVDRM-0286 |
| Description | Membrane proteins are awkward to purify: they are scarce, they sit in detergent, and ordinary resins lose much of them to non-specific binding. This resin addresses that by coupling a mouse IgG2b monoclonal antibody to four percent highly cross-linked agarose, so capture is specific even when the target is present at low abundance, and non-specific binding stays low. Capacity is at least 1.1 mg of protein per millilitre of settled resin. Elution is deliberately gentle: the competing Flag peptide releases the native protein under near-physiological conditions, which matters when the purified material is destined for functional assays or structural work. The antibody recognises the DYKDDDDK epitope whether it sits at the N-terminus, at the C-terminus, or at an N-terminus bearing an additional methionine, so both orientations of construct can be used. |
| Intended Use | Purification and immunoprecipitation of Flag-tagged secreted and membrane proteins, including targets expressed at low levels. |
| Principle / Technology | An immobilised monoclonal antibody binds the DYKDDDDK epitope; the target is later released by competition with excess soluble Flag peptide rather than by denaturing conditions. |
| Sample Type | Clarified cell culture supernatant, lysates or detergent-solubilised membrane fractions containing Flag-tagged protein. |
| Performance Range / Specifications | Capacity at least 1.1 mg of protein per millilitre of resin; low non-specific binding thanks to the specific monoclonal clone and highly cross-linked agarose base; gentle competitive elution; recognises met-Form N-terminal Flag constructs as well as standard N-terminal and C-terminal fusions. |
| Sensitivity / LOD | Does not apply; sensitivity comes from the downstream detection chemistry. |
| Specificity | Mouse IgG2b clone 3F5 coupled to four percent highly cross-linked agarose; capacity at least 1.1 mg protein per millilitre of resin; supplied at 1 mL, 5 mL, 25 mL and 100 mL of settled resin volume. |
| Reaction Conditions / Protocol | Equilibrate the resin, load the clarified sample, wash with Tris-buffered saline, elute with Flag peptide in Tris-buffered saline, then remove the peptide from the eluate as needed. |
| Components / Formulation | Settled resin stored in protective solution; the stated volume refers to the resin itself. |
| Storage Conditions | Store at -25 to -15 °C. Do not freeze in a glycerol-free solution. |
| Shelf Life | 1 year from receipt when stored as recommended. |
| Package Specifications | Supplied at 1 mL, 5 mL, 25 mL or 100 mL of resin. |
| Product Form | Suspension of settled agarose resin in protective solution. |
| Quality Control | Each lot is checked for binding capacity per millilitre and for clone identity. |
| Key Features | - At least 1.1 mg of target per millilitre of resin - Clone 3F5 selected for membrane protein work - Gentle Flag peptide elution keeps native structure - Recognises N-terminal, C-terminal and met-modified N-terminal Flag fusions - Available from 1 mL up to 100 mL of resin volume |
| Purity | Monoclonal antibody of mouse IgG2b class coupled to agarose. |
| Concentration | Binding capacity at least 1.1 mg protein per millilitre of resin. |
| Activity / Unit Definition | Binding performance rather than catalytic units; measured as retained target per millilitre. |
| Source / Origin | Murine monoclonal antibody produced in hybridoma and immobilised on agarose. |
| pH Range / Optimal pH | Protective solution is Tris-buffered saline with fifty percent glycerol at pH 7.4 containing 0.02 percent sodium azide. |
| Shipping Conditions | Shipped cold. |
| Expiration Date / Stability | Stable across the stated shelf life when held at the recommended temperature and kept sealed. |
| Regulatory / Compliance | Supplied for laboratory research purposes. |
| Compatibility | Compatible with Flag peptide elution and suited to subsequent activity assays or structural analysis, since the protein is not denatured during release. |
| Recommended Buffer System | Tris-buffered saline, fifty percent glycerol, pH 7.4, with 0.02 percent mass per volume sodium azide as preservative. |
| Application Notes / Precautions | Never freeze the resin outside its glycerol-containing storage solution, allow the suspension to reach room temperature before opening, and check that azide is removed before contact with living cells. |
| Batch-to-Batch Consistency | Each production lot is released against fixed acceptance criteria so performance stays consistent between orders. |
| Model Type | Immunoaffinity capture resin |
| Starting Material / Tissue Site | Murine IgG2b monoclonal antibody immobilised on cross-linked agarose |
| Delivery Standard | Bottle of resin suspension in protective solution |
| Turnaround Time | No lead time is published on the product page; availability is confirmed when the order is placed. |
| Readouts / Downstream Assays | Yield and purity of the Flag-tagged target after peptide elution, judged by SDS-PAGE. |
For in vitro diagnostic use.
